Yesterday i was going to use my 500D but when i turn it on the card activity light stays on for much longer than usual,
when the screen comes on it says: "card cannot be accessed. reinsert/change the card or format card with camera".
when i try to format the card with the camera i get a message that says it cant format it and to change the card.

the SD card does not work in the cardreaders in any of my computers either.
the card also hasn't been out of the camera since i got the camera in october last year, but i did try to clean the contacts on the card to no avail.

so is it possible for a memorycard to die overnight for no obvious reason?
it was a Transcend SDHC 8GB class 6 card.

when flash memory dies it just dies. check the contact of the card to see if they're not worn out if they are you might be able to save it, if the flash memory died, you are screwed AFAIK.
